Protein Analysis on the 2100 Bioanalyzer

Protein analyses are often required for a range of research applications including structural biology, proteomic analyses, target validation studies, assay development, and high-throughput screening. Typically, researchers use manual techniques like polyacrylamide gel electrophoresis or colorimetric analysis for protein assessment and quantification, however, these techniques are extremely rate limiting and time- or labor-intensive. From expression and purification of recombinant proteins to stability studies and assessment of antibody quality, Agilent offers the Protein 230 and Protein 80 Series II kits to analyze a wide range of samples using lab-on-a-chip technology. When used in conjunction with the Bioanalyzer 2100, each chip is capable of providing size, purity and concentration information for ten different protein samples, all while providing real-time data acquisition. Improvements in newly released kits offer extended size range compatibility, exceptional purity and stability of buffers, higher sensitivity, and reproducible baselines.

The ladder consisting of six recombinant proteins and an upper and lower marker is used for the sizing of unknown protein samples. The gel-like image and the electropherogram are shown.

Samples containing various concentrations of BSA or CAII were analyzed using the Series II Protein 230 assay. The peak height is plotted against the peak size to determine sizing accuracy and reproducibility, independently of the protein concentration.

The analysis of a blank sample (PBS) demonstrates the purity of the sample buffer. There are no detectable impurities in the electropherogram of the Protein 230 sample buffer. The complete electropherogram is shown in A and a baseline zoom in B.

Samples containing 6 ng/µL CAII in PBS were analyzed using the Series II Protein 230 assay. The full electropherogram is shown in A, and the enlargement of the CA II peak in B (S/N ~ 6).
