Agilent continues to provide dependable workflow components to simplify your experimentation. In collaboration with Tecan, Agilent offers compatibility of Agilent's CGH 244K, 2x105K, and now 4x44K microarrays with the Tecan HS Pro Series Hybridization station. The Tecan HS Pro series of hybridization stations provide user-friendly, consistent, and reliable results from sensitive microarray experiments. The HS 4800 Pro and HS 400 Pro Hybridization Stations represent the very latest technology for performing fully automated, highly sensitive and reproducible microarray processing. The systems are designed for full automation, from pre-hybridization to automatic slide drying with OSND™ technology. Tecan's unique ABS (Active Bubble Suppression) system effectively minimizes the risk of bubble formation during a hybridization step, eliminating hybridization artifacts and enhancing experimental consistency. Liquid agitation during hybridization increases sensitivity, stringency and uniformity, and enables reduction of hybridization times.

The Tecan HS Pro offers three types of hybridization chambers, each matching an Agilent microarray format, that allow users to conveniently process different microarray formats based on their experimental needs. Tecan's latest development, the Quadchamber, developed to accommodate Agilent's 4x44K multiplex format, enables customers to achieve process automation of this popular microarray format. When used with Agilent's prehybridization buffer, Agilent's wash buffer and wash buffer additive, the Tecan hybridization station is able to process Agilent microarrays and achieve comparable data while eliminating the manual washing and drying steps in the workflow. The following figures represent a comparison between the Tecan hybridization instrument and the manual "Surehyb" workflow.

Figure 1. Interarray reproducibility for 244K arrays.

Log ratio standard deviations are shown for four replicate arrays hybridized on the Surehyb and Tecan HS Pro platforms. Using the automated Tecan system, the interarray LRSD's decrease from 0.08 with the manual process to below 0.05.

Figure 2. Median intra-array CV for replicated probes on the 244K array.

24-244K arrays hybridized with male/female or HT29/male targets on the Tecan HS Pro instrument exhibit consistently low intra-array CV's of the processed signals. Median intra-array CV's of the replicated probes on these arrays is 3.4% in the green channel and 3.3% in the red channel.

Figure 3. Chromosome and zoom-in views using CGH Analytics.

12-244k arrays hybridized with HT29/XY on the Tecan HS Pro Hybridization Station are plotted in Agilent's CGH Analytics software. The chromosome view plot shows Chromosome 16, while the zoomed-in view focuses on an aberrant region. The 12 arrays, shown with a 3-point moving average, demonstrate consistency in log ratio and the aberration calls.
