Agilent's SureHyb hybridization chamber is optimally designed to integrate with our hybridization oven, enabling more robust and reproducible results for heightened gene detection, improved data quality, and increased sensitivity. The improved SureHyb protocol for hybridization utilizes a bubble to mix the hybridization reagent throughout the incubation process, offering significant advantages over its predecessor, the Legacy 6-screw model. The synergy between the hybridization chamber and oven lies in consistent rotation, enabling the bubble to rotate freely inside the chamber, promoting microarray signal uniformity.
Customer publications demonstrate the improvements in the SureHyb system, particularly with respect to gene detection and reproducibility of results.

Eight microarrays hybridized to both chamber types were analyzed using Rosetta ResolverĀ®. At the 99% confidence level, the SureHyb chamber detected 5,452 ORFs compared to only 5,215 ORFs for the legacy chamber. There were 5,136 common ORFs between the two with zero anticorrelated, suggesting no change in the ORFs log ratio. Thus, the new hybridization tool resulted in 316 unique signature ORFs (green) as compared to only 79 unique ORFs (red) for the legacy tool.

12 microarray self vs. self experiments where six microarrays were hybridized each to either the SureHyb or Legacy hybridization chambers, the CV% values were compared. Plots show a clear difference between the two chamber systems, indicating a two-fold reduction in the mode and median CV% values (mode CV 15% Legacy, 7% SureHyb; median CV 14.5% Legacy, 7.4% SureHyb).
